ILC3s select microbiota-specific T regcells to establish tolerance in the gut

Abstract Microbial colonization of the mammalian intestine elicits inflammatory or tolerogenic T cell responses, but mechanisms controlling these distinct outcomes remain poorly understood, and accumulating evidence indicates that aberrant immunity to intestinal microbiota is causally associated with infectious, malignant diseases. Here we define a critical pathway fate versus cells respond express transcription factor RORγt. We profiled all RORγt +immune at single-cell resolution from intestine-draining lymph nodes mice reveal dominant presence regulatory (T reg) lymphoid tissue inducer-like group 3 innate (ILC3s), which co-localize interfollicular regions. These ILC3s are having interconverting potential extrathymic AIRE-expressing cells, abundantly major histocompatibility complex class II (MHCII), necessary sufficient promote microbiota-specific +T regcells prevent their expansion as helper 17 H17) cells. This occurs through ILC3-mediated antigen presentation, αV integrin competition for interleukin-2. Finally, analyses suggest interactions between impaired in bowel disease.
